چکیده
Psoriasis is an immune-mediated skin condition characterized by hyperproliferation of keratinocytes. stem cells (SCs) have previously been shown to exhibit increased asymmetric SC self-renewal without a change in number. To better understand the genes modulating and committed progenitor cell (CP) proliferation psoriasis, we reexamined Cheng et al. 2018 scRNA sequencing data comparing truncal samples (n=3) from psoriatic normal human epidermis. Focusing on mitotic types, utilized unbiased subclustering approach with pseudotime analysis isolate four clusters corresponding putative SCs CPs. All expressing differentiated markers (IVL FLG) or for melanocytes immune were discarded order keratinocytes basal lower spinous layers. We found that number CPs eight-fold psoriasis no detected layer. As expected, differentially upregulated all However, also identify highly expressed specific highlight their possible role disease pathogenesis including, POUF51 (upregulated SCs), regulator maintenance HES1 CPs), Notch target gene involved keratinocyte division. Understanding basis CP may lead novel therapeutic targets manifestations patients mild moderate disease.

A Graph-Based Clustering Approach to Identify Cell Populations in Single-Cell RNA Sequencing Data
Introduction: The emergence of single-cell RNA-sequencing (scRNA-seq) technology has provided new information about the structure of cells, and provided data with very high resolution of the expression of different genes for each cell at a single time. One of the main uses of scRNA-seq is data clustering based on expressed genes, which sometimes leads to the detection of rare cell populations. ...

متن کاملI-13: Transcriptome Dynamics of Human and Mouse Preimplantation Embryos Revealed by Single Cell RNA-Sequencing
Background: Mammalian preimplantation development is a complex process involving dramatic changes in the transcriptional architecture. However, it is still unclear about the crucial transcriptional network and key hub genes that regulate the proceeding of preimplantation embryos. Materials and Methods: Through single-cell RNAsequencing (RNA-seq) of both human and mouse preimplantation embryos, ...
متن کاملSingle-cell RNA sequencing reveals molecular and functional platelet bias of aged haematopoietic stem cells.
Aged haematopoietic stem cells (HSCs) generate more myeloid cells and fewer lymphoid cells compared with young HSCs, contributing to decreased adaptive immunity in aged individuals. However, it is not known how intrinsic changes to HSCs and shifts in the balance between biased HSC subsets each contribute to the altered lineage output. Here, by analysing HSC transcriptomes and HSC function at th...
متن کاملSequencing Single Cells Reveals Sequential Stem Cell States.
Understanding the molecular mechanisms underlying activation of quiescent neural stem cells (NSCs) is complicated by heterogeneity in coexisting NSC pools. Two papers in this issue of Cell Stem Cell (Llorens-Bobadilla et al., 2015; Shin et al., 2015) report sequencing of single NSCs, providing insights into the transition from quiescence to activation and highlighting common themes in NSCs from...
